Target Information
In mice, the Farnesoid X Receptor (FXR, NR1H4) performs several crucial functions, including regulating bile acid, lipid, and glucose metabolism, and influencing liver and intestinal health. FXR is a nuclear receptor activated by bile acids, predominantly expressed in the liver and small intestine, with its primary role being to maintain the homeostasis of bile acids, cholesterol, and lipids. For instance, mice lacking FXR exhibit increased plasma high-density lipoprotein cholesterol (HDL-C) levels, significantly reduced clearance of cholesterol esters, and elevated levels of non-esterified cholesterol and triglycerides. Additionally, FXR indirectly influences lipoprotein metabolism by regulating the expression of the phosphatidylethanolamine transporter (PEPT) gene. FXR-deficient mice display traits such as insulin resistance, decreased glucose tolerance, and fatty liver.
