Target Information

Tumor necrosis factor receptor superfamily member 6 (also known as Fas) serves as a receptor for TNFSF6/FASLG. Upon activation, the adapter molecule FADD recruits caspase CASP8 to the receptor, forming the death-inducing signaling complex (DISC). This complex facilitates the proteolytic activation of CASP8, initiating a cascade of caspases (aspartate-specific cysteine proteases) that mediate apoptosis. Fas-mediated apoptosis is implicated in the induction of peripheral tolerance and the antigen-stimulated suicide of mature T-cells. In mice, Fas has extensive and complex roles, including immune regulation, cell apoptosis, metabolic regulation, tumorigenesis, and the inflammatory response. Mice deficient in Fas are more susceptible to tumor metastasis because Fas-positive tumor cells can evade clearance by the immune system.
