Target Information
F-box/LRR-repeat protein 3 functions as the substrate-recognition component of the SCF(FBXL3) E3 ubiquitin ligase complex, which is integral to circadian rhythm regulation. It is essential for maintaining both the speed and robustness of circadian clock oscillations. In mice, Fbxl3 primarily regulates the length and stability of circadian rhythms by controlling the degradation of CRY proteins, which in turn influences related behavioral and cognitive functions. Fbxl3 is central to mice's circadian rhythms, and its mutation leads to an extension of the circadian cycle. For instance, the Afh (after hours) mutation involves the substitution of Cys358Ser in Fbxl3, which slows the degradation of CRY1 and CRY2, thereby stabilizing these proteins and extending the circadian cycle from the normal 24 hours to about 27 hours. In mice lacking Fbxl3, the circadian cycle is significantly extended to approximately 29 hours, highlighting Fbxl3's crucial role in regulating circadian rhythm cycle length.
