Eukaryotic translation initiation factor 2-alpha kinase 3 is a protein kinase that senses metabolic stress by phosphorylating the alpha subunit of eukaryotic translation initiation factor 2 (EIF2S1/eIF-2-alpha) in response to various stressors, such as the unfolded protein response (UPR). It is a crucial component of the integrated stress response (ISR) to unfolded proteins: EIF2AK3/PERK specifically identifies and binds to misfolded proteins, triggering its activation and subsequent EIF2S1/eIF-2-alpha phosphorylation. In mice, EIF2AK3 performs a wide range of functions, including mediating the endoplasmic reticulum stress response, regulating insulin secretion, managing metabolic processes, providing neuroprotection, and suppressing tumors.
