Target Information

Ephrin type-B receptor 3 (EphB3) is a receptor tyrosine kinase that interacts promiscuously with transmembrane ephrin-B family ligands on neighboring cells, initiating contact-dependent bidirectional signaling. The signaling pathway activated downstream of the receptor is termed forward signaling, while the pathway activated downstream of the ephrin ligand is known as reverse signaling. In mice, EphB3 plays critical roles in various processes, including the negative regulation of neural stem cell proliferation, inhibition of bone formation, neuroprotection following spinal cord and brain injuries, and promotion of certain cancers. During mouse brain development, EphB3 exerts an inhibitory effect on the proliferation of neural stem cells (NSPCs), particularly in the early stages of hypothalamic development. In EphB3-deficient mice, the proliferation of NSPCs is markedly increased, demonstrating that EphB3 normally acts to maintain balance by negatively regulating NSPC proliferation.
