In mice, the function of ELA1 (trypsin enhancer 1) is primarily associated with the transcriptional regulation specific to pancreatic exocrine cells. The ELA1 enhancer is crucial for regulating gene expression in these cells, utilizing the synergistic action of multiple key components. It comprises three main functional elements: A, B, and C. The A element serves as the binding site for PTF1 (pancreatic-specific transcription factor 1) and is responsible for the specific transcriptional activation of pancreatic exocrine cells. The B element enhances transcriptional activity further by binding to the PDX1·PBX1b·MEIS2 trimer complex, which requires an intact trimer structure for functionality, as the absence of any component significantly reduces its activity. The C element boosts the activity of the A or B element without compromising the specificity for pancreatic cells.
