Target Information

E3 ubiquitin-protein ligase CBL-B functions as an E3 ubiquitin-protein ligase that transfers ubiquitin from specific E2 ubiquitin-conjugating enzymes to substrates, generally leading to their degradation by the proteasome. It acts as a negative regulator for the signal transduction pathways of the T-cell receptor (TCR), B-cell receptor (BCR), and high-affinity immunoglobulin epsilon receptor (FCER1). In naive T-cells, CBL-B inhibits the activation of VAV1 following TCR engagement and enforces a requirement for CD28 costimulation for both proliferation and IL-2 production. In mice, this protein is encoded by the Cblb gene.
