Target Information
Dystrophin serves as a link between the extracellular matrix and the cytoskeleton by anchoring to F-actin and acts as a ligand for dystroglycan. It is a crucial component of the dystrophin-associated glycoprotein complex, which accumulates at the neuromuscular junction (NMJ) and various synapses in both the peripheral and central nervous systems, providing structural stability to the sarcolemma. Additionally, dystrophin is involved in signaling events and synaptic transmission. In skeletal muscle, the absence of dystrophin leads to sarcolemma instability, resulting in muscle damage and dysfunction. For instance, in the mdx mouse model-an animal model for Duchenne muscular dystrophy-a mutation in the dystrophin gene results in dysfunctional dystrophin, causing sarcolemma disruption and pathological changes such as fibrosis and the replacement of muscle with adipose tissue.
