Target Information
Dystrophin Associated Glycoprotein 1 (DAG1) is part of the dystroglycan complex, which plays roles in processes such as laminin and basement membrane assembly, sarcolemmal stability, cell survival, peripheral nerve myelination, nodal structure, cell migration, and epithelial polarization. In mice, DAG1 primarily helps maintain the connection between the muscle cytoskeleton and the extracellular matrix by participating in the dystrophin-glycoprotein complex (DGC), thereby protecting muscles from mechanical damage and preserving their functional integrity. DAG1 is initially synthesized as a single protein and is then cleaved into two components: α-dystroglycan at the N-terminus and β-dystroglycan at the C-terminus. β-dystroglycan anchors to the cytoskeleton through its interaction with dystrophin, while α-dystroglycan binds to extracellular matrix components such as laminin, agrin, and perlecan. This structure allows DAG1 to play a crucial role in maintaining the stability of muscle cell membranes and preventing mechanical damage.
