Target Information
Serine/threonine-protein kinase DCLK1 is a probable kinase implicated in a calcium-signaling pathway that regulates neuronal migration during brain development. It may also play a role in the functions of the mature nervous system. In mice, DCLK1 serves as a critical marker of intestinal stem cells, particularly in the ApcMin/+ mouse model, which is widely used to study colorectal cancer. Research has demonstrated that DCLK1 expression is significantly elevated in intestinal epithelial cells, especially in cancer stem cells, where it is closely associated with tumor proliferation, self-renewal, and epithelial-mesenchymal transition (EMT). Knocking out DCLK1 has been shown to markedly reduce the formation of intestinal adenomas and adenocarcinomas, while also decreasing the expression levels of cancer stem cell markers such as Lgr5, Bmi1, and Msi1. These findings highlight the pivotal role of DCLK1 in intestinal stem cell biology and tumorigenesis.
