DNA damage-inducible transcript 3 protein (DDIT3) is a multifunctional transcription factor involved in the endoplasmic reticulum (ER) stress response. It is crucial in managing various cell stresses and can induce cell cycle arrest and apoptosis when ER stress occurs. In mice, DDIT3 is involved in several processes, such as cell stress response, metabolic regulation, bone formation, pancreatic cancer development, and protection of retinal neurons. DDIT3 is particularly important in managing cell stress, especially under conditions of amino acid deprivation and glutamine deficiency. Research has demonstrated that DDIT3 regulates the balance between triglycerides and oxidative phosphorylation via the GCN2-ATF4 pathway, thereby prolonging the survival of cancer cells.
