Target Information
Diacylglycerol lipase-alpha is a serine hydrolase that specifically hydrolyzes arachidonic acid-esterified diacylglycerols (DAGs) to produce the primary endocannabinoid, 2-arachidonoylglycerol (2-AG). It preferentially cleaves sn-1 fatty acids from DAGs that have arachidonic acid esterified at the sn-2 position to synthesize 2-AG, with minimal activity toward other lipids like monoacylglycerols and phospholipids. A deficiency in DAGLA results in a significant reduction of 2-AG levels in the brain, impacting the regulation of neurotransmitters and synaptic signaling. Consequently, mice lacking DAGLA display anxiety and depressive behaviors linked to decreased 2-AG levels. Moreover, DAGLA is present in neural stem cells and progenitor cells within the dentate gyrus of the hippocampus, and its deficiency significantly hinders the proliferation and differentiation of these cells, leading to impaired neurogenesis in the adult hippocampus.
