Target Information

D-aspartate oxidase specifically catalyzes the oxidative deamination of acidic amino acids, reducing D-aspartate levels in the brain-an amino acid that can function as an agonist for glutamate receptors. DDO is predominantly found in neuroendocrine tissues, like the pituitary gland and adrenal medulla, where its primary role is to degrade D-aspartate, maintaining its balance. Research indicates that Ddo knockout mice (Ddo−/−) exhibit significantly elevated D-aspartate levels, alongside a marked decrease in the synthesis of cortisol and α-MSH (melanocyte-stimulating hormone), which leads to reduced melanocyte-stimulating hormone-related behaviors. D-aspartate influences behavioral and metabolic functions by modulating POMC (pro-opiomelanocortin) biosynthesis. For instance, Ddo−/− mice show significantly reduced POMC expression, correlating with lower α-MSH levels, which in turn affects behavioral performance.
