Target Information

Cytotoxic T-lymphocyte protein 4 (CTLA-4) is an inhibitory receptor that serves as a major negative regulator of T-cell responses. Its affinity for its natural B7 family ligands, CD80 and CD86, is significantly stronger than that of the stimulatory coreceptor CD28. CTLA-4 inhibits T cell activation by competitively binding to B7-1 and B7-2, thus blocking CD28 signaling. This inhibitory effect persists even in CD28-deficient mice, indicating that CTLA-4 functions independently of CD28 signaling. The absence of CTLA-4 results in severe lymphoproliferative disease in mice, characterized by T cell infiltration and tissue damage across multiple organs. Additionally, the role of CTLA-4 inhibition in neurodegenerative diseases has been explored. The loss or dysfunction of CTLA-4 can lead to neurodegenerative diseases, such as autoimmune encephalitis.
