Cytochrome c oxidase subunit 1 is a vital component of cytochrome c oxidase, the final enzyme in the mitochondrial electron transport chain responsible for driving oxidative phosphorylation. This respiratory chain comprises three multisubunit complexes: succinate dehydrogenase (complex II, CII), ubiquinol-cytochrome c oxidoreductase (cytochrome b-c1 complex, complex III, CIII), and cytochrome c oxidase (complex IV, CIV). These complexes work together to transfer electrons from NADH and succinate to molecular oxygen, generating an electrochemical gradient across the inner membrane that facilitates transmembrane transport and ATP synthesis. In mouse models, mutations or abnormal expression of subunit I are linked to various diseases. For instance, in the MRL/lpr lupus mouse model, subunit I expression is significantly reduced, potentially contributing to oxidative stress and immune dysfunction. Furthermore, mutations in subunit I are also associated with the onset and progression of diseases such as neurodegenerative disorders, cardiomyopathy, and cancer.
