G1/S-specific cyclin-D1 functions as a regulatory component of the cyclin D1-CDK4 (DC) complex, phosphorylating and inhibiting members of the retinoblastoma (RB) protein family, including RB1, to regulate the cell cycle during the G1/S transition. Phosphorylating RB1 facilitates the dissociation of the transcription factor E2F from the RB/E2F complex, leading to the transcription of E2F target genes necessary for G1 phase progression. Cyclin D1-CDK4 complexes serve as key integrators of various mitogenic and antimitogenic signals. In mice, G1/S-specific cyclin-D1 (CCND1) is primarily involved in regulating the cell cycle, particularly during the transition from G1 to S phase. The absence of cyclin-D1 in mice results in abnormal tissue and cell development, such as defects in eye and mammary gland development, highlighting its essential role in the normal development of these tissues.
