Target Information

Claudin-3 is crucial for tight junction-specific sealing of the intercellular space, operating through calcium-independent cell-adhesion activity. In mice, claudin-3 serves several vital roles, including maintaining liver metabolism and bile secretion, protecting the intestinal barrier, contributing to the brain barrier formation, and regulating tumorigenesis. Deficiency of claudin-3 can cause liver metabolic disorders in mice, evidenced by the downregulation of lipid metabolism pathways, reduced lipid content, and decreased secretion capacity of primary bile acids. Additionally, the absence of claudin-3 affects bile acid retention and alters bile color, emphasizing its significance in cholestasis and liver fibrosis. The loss of claudin-3 also increases intestinal permeability, impairs mucosal barrier function, and promotes inflammatory responses as well as tumorigenesis.
