Claudins are fundamental components of tight junction complexes that regulate epithelial permeability. While certain claudin family members are crucial for forming impermeable barriers, others facilitate the passage of ions and small molecules. Typically, multiple claudin family members are coexpressed and interact, collectively influencing overall permeability. Claudin-1, in particular, is vital for the integrity of the mammalian epidermal barrier. Research has demonstrated that Claudin-1-deficient mice (Cln1−/−) suffer from severe dehydration and die shortly after birth due to a compromised epidermal barrier, despite having a normal layered structure of keratinocytes. This underscores the essential role of Claudin-1 in maintaining epidermal barrier integrity.
