Target Information
Chromatin assembly factor 1 subunit B is part of the histone chaperone complex, chromatin assembly factor 1 (CAF-1), responsible for assembling histone octamers onto DNA during replication and repair. CAF-1 initiates nucleosome assembly by delivering newly synthesized histones H3 and H4 to replicating DNA, while histones H2A/H2B bind to this chromatin precursor after DNA replication to complete the histone octamer. In mice, CHAF1B is involved in processes such as hematopoiesis, immunity, tumorigenesis, and epigenetic regulation. Research indicates that the absence of CHAF1B in mice leads to the depletion of hematopoietic stem and precursor cells (HSPCs) in the bone marrow, illustrating its critical role in maintaining normal hematopoietic function. CHAF1B is significantly connected to the development and prognosis of various cancers. For instance, in a hepatocellular carcinoma (HCC) model, CHAF1B knockout can inhibit tumor migration, while in lung cancer, CHAF1B enhances cisplatin resistance by promoting NCOR2 degradation. Furthermore, high CHAF1B expression is linked to poor prognosis in several cancers, including non-small cell lung cancer and cervical cancer.
