Muscarinic acetylcholine receptor M3 mediates various cellular responses, such as the inhibition of adenylate cyclase, the breakdown of phosphoinositides, and the modulation of potassium channels through G proteins. In mice, Chrm3 plays extensive and complex roles across multiple systems, including the pancreas, intestine, cancer, and cardiovascular systems. In mice infected with Nippostrongylus brasiliensis, Chrm3 deficiency results in increased intestinal barrier permeability and a weakened mucosal immune response, delaying parasite clearance. In cases of colon cancer and glioblastoma, Chrm3 expression is linked to tumor proliferation and invasiveness. Studies have demonstrated that knocking out or inhibiting Chrm3 can significantly reduce tumor growth and volume.
