M2 receptors, part of the muscarinic acetylcholine receptor family, primarily function through G protein-coupled signal transduction pathways. These receptors are located in the heart, central nervous system, and peripheral tissues, where they help regulate heart rate, temperature control, and anti-nociceptive responses. Moreover, M2 receptors play a role in modulating neuronal excitability. For instance, in the basolateral amygdala, they regulate emotional responses by inhibiting neuronal firing. In mice, M2 muscarinic acetylcholine receptors are involved in various physiological and pathological processes. They serve as inhibitory receptors in the heart, reducing heart rate and contractility by decreasing acetylcholine release. However, in certain pathological conditions, like in diabetic mice, insulin impairs M2 receptor function, resulting in decreased acetylcholine release and gastrointestinal motility disorders.
