Target Information

C-C chemokine receptor type 2 (CCR2) primarily functions as a receptor for CCL2, but it also has the ability to bind to CCL7 and CCL12 chemokines. Its interaction with CCL2 on monocytes and macrophages triggers chemotaxis and migration by activating the PI3K pathway, the small G protein Rac, and promoting lamellipodium protrusion. CCR2 is found on monocytes and macrophages in mice and facilitates the movement of monocytes to sites of inflammation through its interaction with MCP-1. In mouse studies, CCR2-deficient (CCR2-/-) mice displayed significant defects in monocyte migration and were unable to effectively attract monocytes to inflammatory sites, leading to impaired delayed-type hypersensitivity reactions and a decrease in Th1 cytokine production, such as interferon γ.
