Target Information

Protein cereblon acts as a substrate recognition component of the DCX (DDB1-CUL4-X-box) E3 protein ligase complex, which facilitates the ubiquitination and subsequent proteasomal degradation of target proteins like MEIS2, ILF2, and GLUL. The proper degradation of these regulatory proteins is crucial for normal limb development and the expression of fibroblast growth factor FGF8. Loss of the CRBN gene in mice leads to learning and memory deficits, particularly in hippocampus-dependent tasks, mirroring the phenotype observed in human non-syndromic intellectual disability (ARNSID). Research indicates that CRBN influences synaptic transmission and the stability of neuronal synaptic connections by regulating the AMPK-MTOR signaling pathway. Additionally, CRBN supports synaptic function and cognitive abilities by modulating large-conductance calcium-activated potassium channels (BKCa).
