Catechol O-methyltransferase (COMT) is an enzyme that catalyzes the O-methylation of catecholamine neurotransmitters and catechol hormones, leading to their inactivation. It also reduces the biological half-lives of certain neuroactive drugs, such as L-DOPA, alpha-methyl DOPA, and isoproterenol. In mice, COMT has complex and diverse functions, influencing dopamine metabolism, neurotransmitter balance, behavior, and cognitive function. The absence or reduced activity of COMT results in elevated dopamine levels, particularly in the prefrontal cortex (PFC), with notable differences between sexes and brain regions. For instance, male COMT-deficient mice exhibit higher dopamine levels, whereas females show lower levels, highlighting the enzyme's role in modulating dopamine dynamics in a sex-specific manner.
