Carnitine O-acetyltransferase facilitates the reversible transfer of acyl groups from carnitine to coenzyme A (CoA), thereby regulating the acyl-CoA/CoA balance. It is essential for the transport of fatty acids for beta-oxidation and is responsible for synthesizing short- and branched-chain acylcarnitines. In mice, the role of CRAT extends beyond maintaining lipid metabolism and energy balance to include regulating insulin signaling and systemic metabolic homeostasis. In obese mouse models, CRAT expression is influenced by diet and metabolic conditions. For instance, the miR-378 family modulates CRAT expression by targeting its mRNA, which affects fatty acid metabolism and insulin sensitivity in obese mice. Reduced CRAT activity is linked to the accumulation of long-chain acylcarnitines (LCACs) in muscle, highlighting its crucial role in fatty acid metabolism.
