Target Information
Cannabinoid receptor 2 is a heterotrimeric G protein-coupled receptor for the endocannabinoid 2-arachidonoylglycerol, which mediates the inhibition of adenylate cyclase. It may play a role in inflammatory response, nociceptive transmission, and bone homeostasis. In mice, CB2 receptors are found in the brain and are expressed in striatal dopamine neurons, where their activation can inhibit neuron discharge activity, affecting dopamine-related neurobehaviors and addiction behaviors, such as cocaine self-administration. CB2 receptors are also crucial in the tumor microenvironment. In non-small cell lung cancer, the absence of CB2 impairs the anti-tumor activity of CD8+ T cells and NK cells, thus promoting tumor growth. In liver fibrosis, the loss of CB2 leads to increased inflammation, whereas CB2 activation has an anti-fibrotic effect, reducing fibrosis by inhibiting the TGF-β signaling pathway.
