Target Information
Calpain small subunit 1 serves as a regulatory component of the calcium-regulated, non-lysosomal thiol-protease that facilitates limited proteolysis of substrates associated with cytoskeletal remodeling and signal transduction, and it is crucial for embryonic development. As a co-regulator, CAPNS1 works alongside the large catalytic subunits, CAPN1 and CAPN2, ensuring their stability and activity. A deficiency in CAPNS1 leads to the impaired function of both CAPN1 and CAPN2, thereby impacting various biological processes. Conditional knockout (cKO) mice lacking CAPNS1 display hippocampal-dependent memory impairments, such as reduced freezing behavior in fear conditioning tests and difficulties in short-term memory formation. Additionally, CAPNS1 deficiency results in presynaptic malfunctions, including decreased synaptic transmission amplitude and accumulation of synaptic vesicles.
