Target Information

Calpain-1 is a calcium-regulated non-lysosomal thiol-protease that facilitates limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction. In mice, CAPN1 has functions across various areas such as cardiomyocyte death, neurodegenerative diseases, liver damage, skeletal muscle function, platelet aggregation, cancer progression, and genetic disorders. As a calcium-dependent protease, it plays a critical role in many physiological and pathological processes. In particular, CAPN1 is instrumental in cardiomyocyte death. Research indicates that exercise-induced calcium influx can result in calcium overload in cardiomyocytes, activating CAPN1. This activation leads to cardiomyocyte apoptosis and arrhythmic cardiomyopathy (ACM) by cleaving apoptosis-inducing factor (AIF) and promoting its translocation to the nucleus. Furthermore, overactivation of CAPN1 is linked to mitochondrial dysfunction and cell death.
