Target Information
Upon ligand binding, bone morphogenetic protein receptor 1A (BMPR1A) forms a receptor complex composed of two type II and two type I transmembrane serine/threonine kinases. In this complex, type II receptors phosphorylate and activate type I receptors, which subsequently autophosphorylate and engage SMAD transcriptional regulators. BMPR1A serves as a receptor for BMP2, BMP4, GDF5, and GDF6, and it enhances chondrocyte differentiation through its interaction with GDF5. In mice, Bmpr1a is involved in various functions including bone development, tumor suppression, tissue repair, and neural and endocrine activities. It plays a crucial role in bone development, as evidenced by the specific knockout of Bmpr1a in osteoblasts, which results in increased bone mass due to the excessive proliferation of osteoblasts and decreased bone resorption. Furthermore, Bmpr1a signaling influences osteoblast differentiation and function via the Wnt/β-catenin pathway.
