Target Information
Beta-secretase 1 is an enzyme that processes the amyloid precursor protein (APP) through proteolytic cleavage. It cuts at the N-terminus of the A-beta peptide sequence, specifically between residues 671 and 672 of APP, resulting in the production and extracellular release of beta-cleaved soluble APP, along with a corresponding cell-associated C-terminal fragment that is later released by gamma-secretase. Research has shown that the loss or inhibition of BACE1 can substantially lower Abeta levels in the brain, thus reducing amyloid plaque formation. For instance, BACE1 knockout mice (BACE1-/-) display significantly reduced Abeta levels while not exhibiting any noticeable developmental defects or behavioral abnormalities.
