Target Information
Ubiquitin-like modifier-activating enzyme ATG7 is an E1-like activating enzyme involved in two ubiquitin-like systems essential for the cytoplasm to vacuole transport (Cvt) pathway and autophagy. It activates ATG12 for its conjugation with ATG5 and the ATG8 family proteins for their conjugation with phosphatidylethanolamine. Mice lacking Atg7 exhibit skeletal dysplasia, reduced bone mineral density, and impaired differentiation of bone marrow cells. These issues are closely tied to defective autophagy in osteoblasts, resulting in increased mitochondrial stress and apoptosis. Additionally, Atg7 deficiency impacts mitochondrial function in skeletal muscle, leading to heightened mitochondrial swelling and increased levels of protein carbonylation.
