Mitochondrial Arginase-2 may play a role in regulating arginine metabolism outside the urea cycle and in reducing nitric oxide synthesis. In extrahepatic tissues, arginase modulates L-arginine availability for nitric oxide synthase (NOS). Arginine metabolism is crucial for regulating both innate and adaptive immune responses. Arginase-2 appears to negatively regulate the survival of activated CD4+ and CD8+ T cells and is important in anti-tumor immunity. Research has demonstrated that mice lacking Arginase-2 (Arg2−/−) show enhanced CD8+ T cell function, evidenced by quicker activation, increased cytotoxicity, and greater production of IFN-γ and IL-2. Additionally, when treated with tumor vaccines, these Arg2−/− mice exhibit significantly slowed tumor growth, indicating a stronger anti-tumor response.
