Target Information
Anti-dsDNA IgG facilitates the breakdown of double-stranded DNA (dsDNA) by binding to it and activating calcium and magnesium ions. Although this process requires energy, the antibody binding lowers the activation energy, thus accelerating the reaction rate. In mouse models, anti-dsDNA IgG has been shown to promote tubulointerstitial fibrosis by inhibiting the suppressor of cytokine signaling 1 (SOCS1). Specifically, they suppress SOCS1 expression and activate the JAK2/STAT1 signaling pathway, which leads to increased production of fibroblast factors such as TGF-β1, PDGF-B, and CTGF, playing a significant role in renal fibrosis. Additionally, anti-dsDNA IgG can bind to self-antigens or foreign antigens, forming immune complexes that deposit in the glomerular basement membrane, triggering the complement cascade and resulting in tubular damage and renal dysfunction.
