Target Information
In mice, Aβ1-42 is primarily associated with neurotoxicity, impairment of synaptic plasticity, and memory dysfunction. While it may exhibit some protective effects at low concentrations, high concentrations of Aβ1-42 oligomers are central to the pathology of Alzheimer's disease (AD). At elevated levels, Aβ1-42 exerts significant toxic effects on neurons. Research has shown that Aβ1-42 oligomers, also known as ADDLs, play a crucial role in inhibiting long-term potentiation (LTP) in the hippocampus, which is closely linked to the loss of synaptic plasticity and memory function. Additionally, these oligomers exacerbate neurotoxicity by activating NMDA receptors and NR2B subunits. In mouse models, Aβ1-42 treatment significantly impairs hippocampal LTP and leads to reduced learning and memory capabilities.
