Alpha-1-antitrypsin (AAT) is a crucial serine protease inhibitor primarily synthesized in the liver and distributed throughout the body via the bloodstream. In mice, α1-antitrypsin plays several roles, including anti-inflammatory actions, immunomodulation, protection against pathogen infections, and safeguarding against tissue damage. It exhibits significant anti-inflammatory properties; research has demonstrated that it can mitigate inflammatory responses by inhibiting the release of inflammatory mediators like tumor necrosis factor α (TNF-α) and interleukin-1β (IL-1β). Additionally, α1-antitrypsin is effective in combating pathogen infections. For instance, in a mouse model of Pseudomonas pneumonia, transgenic mice overexpressing α1-antitrypsin displayed reduced lung inflammation and damage while inhibiting pathogen invasion and preserving the integrity of the epithelial barrier.
