Target Information
Allograft inflammatory factor 1 (Aif1) is an actin-binding protein that enhances membrane ruffling and RAC activation. It boosts the actin-bundling activity of LCP1, binds calcium, and plays a role in RAC signaling and phagocytosis. Aif1 is also implicated in macrophage activation and function, promoting the proliferation of vascular smooth muscle cells and T-lymphocytes, while enhancing lymphocyte migration. Additionally, it contributes to vascular inflammation. In mice, Aif1 has broad and complex functions, spanning immune responses, neurodegenerative diseases, metabolic regulation, vascular diseases, and intestinal microbial immunity. Mice lacking Aif1 exhibit resistance to obesity and insulin resistance induced by a high-fat diet, a phenomenon linked to increased levels of norepinephrine in adipose tissue. Aif1 is also involved in the differentiation and functional regulation of adipocytes, further influencing energy metabolism. These findings highlight the multifaceted role of Aif1 in immune and metabolic processes.
