Target Information

A disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5) is a metalloproteinase critical for connective tissue organization, development, inflammation, and cell migration. As an extracellular matrix (ECM) degrading enzyme, it exhibits proteolytic activity toward the hyalectan group of chondroitin sulfate proteoglycans (CSPGs), including ACAN, VCAN, BCAN, and NCAN, cleaving these proteins at Glu-Xaa recognition motifs. ADAMTS5 is involved in embryonic development, contributing to limb and cardiac morphogenesis and skeletal muscle development through its VCAN remodeling properties. In mice, ADAMTS5 has multiple biological functions, impacting joint health, tissue repair, metabolic regulation, immune response, and the health of the cardiovascular and nervous systems. It is a principal protease responsible for articular cartilage degradation, with its activity closely linked to cartilage damage in arthritis. Research indicates that mice lacking ADAMTS5 experience milder cartilage damage in a surgically induced arthritis model, highlighting ADAMTS5's significant role in the pathological progression of arthritis.
