Target Information
Disintegrin and metalloproteinase domain-containing protein 17 (ADAM17) is responsible for cleaving the membrane-bound precursor of TNF-alpha, converting it into its mature soluble form. It also facilitates the proteolytic release of soluble JAM3 from the surface of endothelial cells and is involved in the proteolytic processing of ACE2. In mice, ADAM17 has a broad range of functions spanning embryonic development, immune regulation, inflammatory responses, cardiovascular disease, metabolic syndrome, and neurodegenerative diseases. It influences chondrocyte differentiation and bone development by regulating the TGF-α signaling pathway. Additionally, during embryonic development, ADAM17 plays a critical role in regulating angiogenesis, and its absence can lead to vascular branching disorders and bleeding.
