Target Information
Rabbit osteonectin (SPARC) is a 32 kDa secreted calcium-binding glycoprotein whose N-terminal acidic module and two EF-hand loops bind collagen I and hydroxyapatite, orchestrating matrix mineralization and cell-matrix adhesion. A solvent-exposed 16-aa "KVE" loop (aa 85-100) between the follistatin and EC domains folds into a β-hairpin unique among rabbit matricellular proteins, presenting a conformational epitope preserved after decalcification and fixation. Affinity-purified antibodies targeting this loop trace native osteonectin in cortical bone, periodontal ligament, and corneal stroma, delivering crisp extracellular fibrils and single-band clarity on immunoblots with no cross-reaction to SPARC-like 1 or thrombospondin.
